The Colosseum Bosses are located within the Colosseum. To access them you must first be flagged by reaching a certain arena in the regular Colosseum. Then simply use their statue to enter their personal chamber. When you kill the boss and any remaining creatures, a door will appear leading back to the Colosseum drop point.

Strategy : to kill her easily, have two melees with olthoi slayers weapon and an imperiler/vulner/healer/stamer. one of the melee should attack the clawbearers first, while the other should concentrate on the matron (use priority settings in Ltxi) with that strategy, the matron will be dead in 5 minutes. (attack her in the center of the room to prevent her from teleporting)
